Uses

  • Multi-drug-resistant tuberculosis (MDR-TB)

Dose and Duration

MDR TB (in combination with other anti-TB drugs)

Adult and child: 15–20 mg/kg (max 1 g) once daily. Can be given in 2 divided doses to improve tolerance

Contra-indications

  • Hypersensitivity to prothionamide
  • Severe liver disease
  • Porphyria

Side Effects

  • Transient elevation of liver function tests
  • Nausea, vomiting, diarrhoea, excessive salivation, metallic taste, stomatitis, abdominal pain
  • Anorexia
  • Acute hepatitis
  • Dizziness, encephalopathy, peripheral neuropathy

Patient Instructions

  • Take your medicine with or after a meal to reduce gastrointestinal adverse effects

Pregnancy

  • Do not use

Breast-feeding

  • Do not use

⚠️ Caution

  • Reduce dose in severe renal impairment
  • Monitor hepatic function once every 3 months during treatment or once every month if used concomitantly with P-aminosalicylic acid
  • Monitor blood glucose in diabetic patients
